'Inderst inde' bliver fortalt af følelserne inde i pigen, Rileys, hovede. Fortælleren er Glæde og hendes venner Vrede, Frygt, Tristhed og Afsky. I mange år har Glæde styret showet, men da Rileys forældre beslutter sig for at flytte til en anden by, forsvinder Glæde og Tristhed fra Rileys hovede. Nu er det Vrede, Frygt og Afskys job at holde styr på Rileys følelser, og det går ikke helt så godt.
Da Anders til sin fødselsdag får robothelten Buzz Lightyear i gave, skaber det forvirring på børneværelset.
