Halvaantunut merijalkaväen sotilas Jake Sully lähetetään Pandoralle suorittamaan haastavaa tehtävää. Koska Pandoran ilmakehä on ihmiselle myrkyllistä, kloonataan Jake avatar-kehoon, joka on sekoitus ihmis-DNA:ta ja planeetan paikallisen navi-heimon DNA:ta. Jake saa kloonauksen myötä myös liikuntakykynsä takaisin. Pandoralla Jake löytää harvinaislaatuisen kauniin, mutta myös vaarallisen maailman. Hän tutustuu viehättävään navi-neitoon, ja suhde tähän muuttaa kaiken. Pikkuhiljaa Jake lunastaa paikkansa navi-yhteisössä ja huomaa kotiutuneensa heidän maailmaansa. Tehtävän päättyessä Jake joutuu vaikeiden valintojen eteen. Hänen täytyy johtaa taistelua ihmiskunnan pelastamiseksi - mutta omiaan vastaan.

Seurue joutuu paikallisten arikara-intiaanien hyökkäyksen kohteeksi ja menettää valtaosan miehistään. Eloonjääneet suuntaavat Glassin johdolla kohti 300 kilometrin päässä sijaitsevaa Fort Lookoutia. Glass joutuu talvisessa metsässä harmaakarhun raatelemaksi ja ryhmää johtava kapteeni Andrew Henry ei usko Glassin selviävän vammoistaan. Henry jättää ryhmästään Bridgerin ja Fitzgeraldin miestä jäämään tämän luokse ja hautaamaan hänet kunniallisesti kun aika koittaa. Miehet päättävät haudata Glassin elävältä ja varastaa hänen aseensa ja varusteensa. Glass jää kuitenkin henkiin ja aloittaa pitkän ja vaikean vaelluksen takaisin elävien kirjoihin ja kostaakseen hänet pettäneille miehille.

Vakavasti sairas Prinssi Ashitaka on matkalla kauas itään etsimään parannusta villisikajumalan langettamaan kiroukseen, mutta joutuu yllättäen keskelle metsän eläinten ja raudan takomiseen erikoistuneen Tataraban klaanin välistä sodankäyntiä. Esiteollinen kaivosyhteisö on vallannut metsää, jota uhkaa kuolema hyväksikäyttäjien kynsissä. Ashitaka kohtaa sotatantereella Sanin eli Prinsessa Mononoken, susien kasvattaman ihmislapsen, joka johtaa metsäneläinten laumoja. Samainen Shishi-Gami on ainoa, joka pystyy vapauttamaan Prinssin kirouksesta. Ashitaka joutuu pulmalliseen välikäteen ja rauhanrakentajan asemaan.
