Myyttiseksi Baba Yagaksi venäläismafian keskuudessa ristitty John Wick luulee pääsevänsä vihdoin viettämään rauhallisia eläkepäiviään autonsa ja uuden lemmikkikoiransa kanssa. Oven taakse ilmestyy kuitenkin vanha tuttu, italialainen gansteri Santino D’Antonio. Santino hyödyntää Wickiin salaista sinettiä. Sinetti on verivala, jonka vahvistamana Wick on jäänyt hänelle velkaa ja Santinon mielestä nyt on tullut aika kuitata maksu suoritetuksi palkkamurhan muodossa. Santinon määräämä keikka vie Wickin Roomaan, jossa pelin henki muuttuu oleellisesti. Herrasmiessäännöillä pelaava Wick huomaa joutuneensa itse maailman parhaiden ammattitappajien iskulistoille.
