엉뚱한 학설을 내세워 망신살이 뻗친 고생물학자 릭 마샬(윌 페렐)은 심기일전, 와신상담을 외치며 학계를 놀라게 할 연구를 위해 정글로 향한다. 마샬과 그의 조수 홀리(안나 프리엘)가 가이드 윌(대니 R. 맥브라이드)의 도움을 받아 강을 거슬러 오르던 중 정체불명의 소용돌이가 일어나고, 물살 속에 열린 시간의 문을 통해 일행은 로스트 시티에 불시착한다.
