De film speelt zich af in twee gescheiden werelden. De eerste is een toekomstige versie van de Aarde waar in november 2012 een gigantisch monster uit de Stille Oceaan opdook en de stad Osaka in Japan aanviel. De andere wereld is de Anteverse, waar het monster vandaan kwam.
De invloedrijke redactrice Margaret dreigt uitgezet te worden naar haar vaderland Canada. Om dit te voorkomen dwingt ze haar nietsvermoedende en komische assistent Andrew om zich met haar te verloven, terwijl ze hem al jarenlang het leven zuur maakt. Hij gaat akkoord met de verloving maar tegen een paar persoonlijke voorwaarden. Het onwaarschijnlijke stel reist af naar Alaska om de eigenzinnige familie van Andrew te ontmoeten.
Nadat hun vliegtuig crasht, strandt een groepje medewerkers van een olieplatform in het ijskoude Alaska. Ze bevinden zich in het territorium van een groep wolven die hen als indringers zien. Ze zullen nu moeten vechten om te overleven om niet ten prooi te vallen aan de hongerige wolven.

In het afgelegen dorpje Nome in Alaska verdwijnen al sinds de jaren zestig onwaarschijnlijk veel mensen. Een officiële verklaring hiervoor is nooit gevonden, maar bijna iedereen in het dorpje lijkt wel te weten waar het door komt: alle vermiste personen zijn ontvoerd door buitenaardse wezens. Sommige dorpelingen zijn ontvoerd en keerden hierna terug in Nome. Zij luchten hierover uitgebreid hun hart bij psychiater Abbey Tyler (Milla Jovovich).
Het meer van Springfield is ongelofelijk vervuild en er moet iets worden gedaan. Iedere bewoner stopt vervolgens met afval in het meer te gooien en het probleem lijkt te zijn opgelost. Maar als Homer Simpson dan de uitwerpselen van zijn net aangeschafte varken in het meer dumpt, gaat alles helemaal mis. Homer en zijn familie weten te ontsnappen en vluchten naar Alaska. Maar daar horen ze dat de regering Springfield geheel van de kaart wil vegen en weten dat ze hun woonplaats moeten zien te redden.
