Syng-gjengen er tilbake i en feelgood-film for hele familien, proppfull av fengende sanger. Den evig optimistiske koalaen Buster Moon samler troppen til deres mest ambisiøse prosjekt så langt: Å sette opp et helt nytt show i storbyen. For å få innpass hos den nådeløse teatermogulen Jimmy Crystal, lover Buster at den tilbaketrukne rockelegenden Clay Calloway skal opptre på scenen. Det viser seg å være lettere sagt enn gjort. Buster og gjengen legger ut på en emosjonell reise mot forsøket på suksess, med stadige påminnelser om musikkens kraft til å lege alle sår.
