Transformers : The Last Knight จะโฟกัสไปที่ ออพติมัส ไพรม์ ออโต้บอตส์ที่กำลังกลับไปยังดาวบ้านเกิดอย่าง ดาวไซเบอร์ตรอน และพบว่าดาวของตนเองได้ถูกทำลายลง ซึ่งเขานั้นก็มีส่วนที่ทำให้ดาวถูกทำลายด้วย เพื่อจะทำให้ดาวของเขากลับมาเป็นปกติสุขได้นั้นออพติมัส ไพรม์ ต้องตามหาวัตถุชิ้นหนึ่งที่อยู่บนโลกมนุษย์ และวัตถุที่ว่านั้นก็เกี่ยวโยงไปถึง “เมอร์ลิน” พ่อมดในตำนานสมัยกษัตริย์อาเธอร์ด้วย แต่ออพติมัสพลาดท่า จนถูก“ผู้สร้าง” (ควินเทชชั่น เผ่าพันธุ์ผู้ทรงภูมิ จุดกำเนิดทรานฟอร์เมอร์ส ตามฉบับการ์ตูน) ทำให้กลายเป็นศัตรูของเหล่ามนุษย์ไป
